Well, a very wise person once told me that soul mates are two people who come together like puzzle pieces. Neither is perfect. In fact, one or both can be deeply flawed. However, their pieces somehow come together and mesh perfectly. If you stand back and examine your own personal relationships and you are honest, you can see this in your own partner. You're each stronger because of the other. You complete each other. I also believe that two people who are too much the same will butt heads. So usually I like my FMC & MMC to be opposites in one way or another. It's always fun to watch them be forced to compromise and meet halfway.
Creating the chemistry is different for each couple. They need to click. Each needs to make a lasting impression on the other, whether good or bad. The impression needs to stick so that days, weeks, maybe even months later they are still bubbling or cursing under their breath about it. I'll take either. ;) Then I build on that. If she wants to hate him, this will now become a challenge. Same goes for him because there will be one very redeeming quality that will suck the other person in. Then again I build on that. Their conversations have to be entertaining whether they're romantic, mysterious (cryptic), or argumentative. There has to be something that, regardless of the topic of conversation, leaves both MCs and the reader wanting and waiting for them to talk to one another again.
All this and their body language go hand in hand: the tense stance when trying hard not to give into to doing or saying what they really want and the deep, intense uncontrollable gazes. The unforgettable ones give them (sometimes infuriatingly) the ever-present-in-romance-novels butterflies.
My husband often asks me as he passes by my desk and I'm writing, "What's that goofy smile about?" That's usually a good sign that the chemistry is working, because if I'm feeling the irrepressible need to smile like a goofball as I'm writing the dialogue, then we're good. LOL. On a sidenote, building to that moment—creating that chemistry that will not be suppressed no matter how hard either side tries—is my favorite part of writing romance. =D
